Research Interests
| Our research group are interested in synaptic transmission and plasticity in the central nervous system and particularly excitatory amino acid neurotransmission (glutamate) and long-term potentiation (LTP) and long-term depression (LTD) in the hippocampus. LTP is an activity dependent enhancement of synaptic transmission in the vertebrate brain, which because of its long duration and association properties, is now widely used for the investigation of cellular mechanisms for memory formation. We are presently concerned with the role of cytokine action on LTP. Currently we are carrying out research to elucidate the modulatory role of cytokines and in particular interleukin-1 and TNF-alpha on LTP using electrophysiological techniques including extra-cellular field recording and whole cell patch clamp recording. Peptides including many of the pro-inflammatory cytokines are known to play a major role in the modulation of synaptic plasticity. We have recently provided evidence for a role of metabotropic glutamate receptors in the TNF alpha -induced inhibition of LTP. Other on-going research in our laboratories includes studies into the effects of mild hypoxia on hippocampal slices. We also have a laboratory in the Chemistry Department in the National University of Ireland, Maynooth, Co Kildare, investigating the real time monitoring of transmitter release (dopamine, noradrenaline and 5-HT) in the CNS using the technique of fast cyclic voltammetry. The group is currently investigating the action of multiple peptides on real time stimulated dopamine release in the rat brain slices containing the striatum and nucleus accumbens.
