Research Interests
| Our research involves the study of kidney fibrosis and end-stage renal disease. The incidence of end stage renal disease (ESRD) is rising at a rate of 8-10% per annum. While the initiating factors of the disease may vary, tubulo-interstitial fibrosis is a keMolecular and Cellular Biology of Renal Fibrosis. Our research involves the study of kidney fibrosis and end-stage renal disease. The incidence of end stage renal disease (ESRD) is rising at a rate of 8-10% per annum. While the initiating factors of the disease may vary, tubulo-interstitial fibrosis is a key event and indicates a likelihood of progression to ESRD. Therapeutic strategies to halt the progression to ESRD are very limited mainly because of a lack of understanding of the mechanisms involved in tubulo-interstitial fibrosis. Molecular and Cellular Biology of Renal Fibrosis We believe that a process known as epithelial-mesenchymal transition (EMT), whereby epithelial cells become more fibroblast-like, may be involved in the development of kidney fibrosis. EMT is a physiological process during embryo development but in the adult can play a fundamental role in the early stages of transformation, invasion and metastasis of cancer cells. This process involves the loss of epithelial cell markers such as E-cadherin and the increase in mesenchymal markers such as alpha-smooth muscle actin. We are using DNA microarray analysis to determine specific gene changes in cell culture models of renal fibrosis, including EMT models. Renal Pharmacology and Toxicology We are also interested in the development of models of renal cell injury, including the investigation of the role of immunosuppressant agents in renal fibrosis through the application of genomics and proteomics techniques. Studies on differentially regulated genes and proteins provide insights not only into immunosuppressant-induced renal fibrosis, but also into the molecular mechanisms underlying EMT in other contexts, including tumour biology and fibrosis of other organs. Further examination of differentially regulated genes may lead to the identification of novel genes and potential adjunct therapies. The major focus of our research is providing new models to develop our understanding of the mechanisms of nephrotoxicity and kidney damage, with the aim of identifying new therapeutic targets and strategies.
