Research Interests

My area of research focuses on circulating anucleate platelets and their precursor megakaryocytic cells in the bone marrow.  Traditionally considered the principle effectors of haemostasis, increasing evidence now suggests that platelets fulfil a much broader role in health and disease, e.g., contributing to bone remodelling and disease, the immune response as well as angiogenesis and tumour growth. 


Our work over the past 10 years has focused on a comprehensive assessment of the anucleate platelet proteome based on a sequential series of analyses that included fractional proteomics (1), characterisation of specific interactomes and more recently detailed descriptions and functional analyses of novel platelet pathways including the discovery of the WNT signalling network in platelets (2) and megakaryocytes (3).  In brief, we found WNT to be one of the few known secreted endogenous regulators of platelet activity which prevents the earliest phases of platelet activation (2). Our experiments in genetically modified mice demonstrated that endogenous WNT acting through the Fzd6 receptor normally dampens platelet reactivity (2), akin to other endogenous negative platelet regulators such as prostacyclin and nitric oxide. Intriguingly, we have recently found that WNT3a strongly promotes proplatelet formation by primary megakaryocytes, an effect which we can be completely attenuated using the canonical inhibitor DKK1 (3). This implicates WNT signalling in the regulation of platelet count, which can greatly influence the risk of thrombosis in patients with coronary artery disease and may play a role in unexplained thrombocytopenia
