Biography

Professor Mohamed Al-Rubeai graduated from the University of Baghdad in 1969 and obtained his PhD in Genetics from QMC, University of London in 1979.  He was the first in the world to elucidate the genetic basis of radiation sensitivity in plants. Immediately after receiving his PhD he joined the University of Garyounis as a lecturer in Biology. He returned to the UK to work on a BBSRC research fellowship at the University of Surrey. His work on purification and characterization of dihydrofolate reductase from Mycobacterium spp. was highly cited. He moved to the University of Birmingham in 1988 where he established himself as a world leader in cell culture technology with his work on shear sensitivity of animal cells in bioreactor and establishing flow cytometry as a major technique for the monitoring and control of cell culture. His most cited papers were written at this time, in which he showed that animal cells in in-viro culture die by apoptosis. His EU Framework 4 project was recognized in 1995 by the EU programme committee as the best research proposal in the area of Biotechnology. He was chosen for the 1991 Royal society visit to Prague after the velvet revolution.  In 1998 the British Council selected him for a lecture tour in Singapore to present British Biotechnology. He was appointed to the Chair of Biotechnology in 2000. He also spent time as an adjunct professor at the University of Queensland, Australia. In 2005 he moved to University College Dublin to take up the newly established Chair of Biochemical Engineering and received more than 2 million euro grant from the SFI. He remained as an adjunct professor at the University of Birmingham until 2008. Professor Al-Rubeai is the editor of the series of "Cell Engineering", reviews editor of "Cytotechnology", and member of the editorial board of "Biotechnology and Applied Biochemistry". He also spent time on the board of "Biotechnology and Bioengineering" and Pharmaceutical Bioprocessing and on the advisory board of Sigma-Aldrich and Vivalis. He acted as an advisor to the Ministry of Higher Education in Iraq, scientific advisor to the President of Iraq, advisor to UNESCO and as a visiting professor and external assessor in University Technology Malaysia. He co-authored "Engineering a Knowledge Island 2020" for the Royal Irish Academy. He was recognized for his contribution to the development of cell culture technology by the Institute of Chemical Engineers, UK with their Donald Medal in 2008. He is Fellow of the Royal Society of Biology. He has authored over 500 professional papers, reviews, chapters and patents that reflect his broad interest in production of biopharmaceuticals, mammalian cell culture, tissue engineering, stem cell bioprocessing, metabolic engineering and biomaterials. He supervised 47 PhD and 51 MSc dissertations, gave more than 50 invited talks in international conferences and chaired several of them, lectured in more than 50 universities, research institutes and biopharmaceutical companies and awarded in excess of 20M Euro as research grants.

Research

Research Interests

The Animal Cell Culture Technology Group conceived with the SFI support in 2005 in the School of Chemical and Bioprocess Engineering and Conway Institute of Biomolecular & Biomedical Research at the University College Dublin is the first and only such group in Ireland dedicated to the process engineering of biotechnological processes involving animal cells based on a combination of engineering, analytical, biological and physiological skills.

The research objectives of the group concern the provision of design bases for the more effective and economic application of intensive production methods for mammalian cells. In particular, the intensification of such processes is dependent upon understanding the physiological determinants of cell growth and death, and of product synthesis and secretion, and also the physical determinants of culture performance in the intensified bioreactor environment.

The group approach to cell culture development and optimisation is to provide an understanding of the relationship between gene and protein expression and growth and productivity. On the basis of this improved understanding novel strategies for optimisation of cell culture should be possible. These strategies include the development of methods to improve cell culture, survival and proliferation of mammalian cells. Underlying these efforts is the need for rapid, reliable techniques for selection of high producers and monitoring cell viability and physiology. Underlying the work its central coherence with existing key research themes within the Conway Institute (e.g. gene array analysis, proteomics and cytomics). The programme is focused exclusively on strategically important themes in the development of mammalian cell culture processes for enhanced and optimised production of biopharmaceuticals. Mammalian cell culture processes face numerous challenges related to compressed product development cycles, capacity shortages, and the proliferation and productivity of cell lines and culture conditions. Exploitation of the advances in molecular biology will help to resolve many of the problems associated with large scale production.

The group is also interested in research in the following areas of cell culture technology:

§         In-line bioprocess monitoring by cytometric methods

Conventional off-line techniques for monitoring culture conditions are time- and labour-intensive, and incompatible with modern development and manufacturing timelines. The development of flow cytometric techniques has greatly advanced our ability to identify and characterise the morphological and biochemical heterogeneity of cell populations, permitting a rapid and sensitive cell analysis. If it is to make a real impact on the control of cell culture processes, however, the technique must become available on-line. This proposal is to eliminate this critical limitations for extensive investigations of single cell parameters. The obtained results have convinced us that the time is ready for the next step of development in single cell analysis that promises significant further advances in studying, monitoring and controlling cell cultures.

Project objectives:

1.  To build a prototype of an automatic sample handling and staining device and interface it with a dedicated bioreactor/flow cytometer system for on-line analysis of cell populations.

2.  To develop protocols for on-line staining of cell populations.

The steps performed automatically will include sample dilution, cell fixation and cell staining to permit quantitation of the macromolecular composition of individual cells. The staining steps will be carried out in a specifically designed microchamber.This work is part of a programme funded EI Bioindustry Led Research Project on bioprocess application of process analytical technology (apPAT) which is based on previously funded project by EI (BioMon), which developed a prototype device for the automated preparation of process samples for cell culture characterisation based on flow cytometric measurements. This project (apPAT) will demonstrate and validate a capability for improved process monitoring and control using Process Analytical Technology (PAT). This project will address the integration of these technologies with a commercial data management and batch monitoring and control system which will provide the platform for the required data collection, analysis and control loop implementation. The project will address the critical issue of aseptic automated sampling through a rigorous evaluation of available sampling systems, with appropriate modification where necessary. It is anticipated that the monitoring protocols developed will provide a basis for routine automated sampling and measurement for cell culture bioreactors.

§         Development of scalable systems for human red blood cell production from heamatopoietic stem cells

Ex vivo expansion of erythroid progenitors from cord blood and other sources is a potential source for supply of blood products which would significantly improve the transfusion capacity of blood centres (Neildez-Nguyen et al. 2002 Nature Biotechnol. 20, 467). While the maintenance of erythropoiesis and enucleation mechanisms in vivo depends on a delicate interaction of different growth factors, lineage specific cytokines and haematopoietic microenvironment, recent work has demonstrated that the primitive haematopoietic stem cells (CD34+) could be expanded and converted ex vivo into mature RBCs (Giarratana et al. 2005 Nature Biotechnol. 23, 69). In vitro generated RBCs have all the characteristics of functional, native, adult RBCs in terms of enzyme content, membrane deformability and the capacity to fix and release oxygen. In this project, we are seeking to develop a continuously perfused free suspension or encapsulated system to provide suitable environment for high growth rate with high potential for fully maturation into RBCs and facilitates long term in vitro growth of haematopoietic stem cells and their differentiation into a pure erythroid lineage.  

§         Cell-nanosurface interactions (Precision Programme).

Precision is a Strategic Research Cluster supported by Science Foundation Ireland and our Industrial partners. The Cluster aims to develop the scientific and technological knowledge needed for present and future manufacturing applications using plasmas, with a specific emphasis on nano-scale products, process reliability, manufacturing costs and advanced materials processing. It enables the fundamental understanding of plasma processing to be coupled with real word product application in the medical device and food packaging industries. As part of this project, our group will investigate: 1) The influence of surface modifications on progenitor cell (bone marrow mesenchymal cells (MSC)) adhesion and differentiation. 2) The deposition of nano-coatings that contain natural antioxidants onto food packaging material.

§         Bioprocessing of virally transduced cells for the application of gene therapy.

§         Process intensification and development of serum free media.

§         Development of a scaleable disposable bioreactor for animal cell culture.

§        Application of systems biology to understanding the behaviour of mammalian cells and enhancing their targeted productivity.

§            Expansion of Chondroprogenitor cells and development of 3D engineered cartilage
