Protein biochemistry techniques including 2D-electrophoresis for proteome analysis, protein fractionation and protein purification methods. Separation of protein mixtures using HPLC. MS analysis of proteins and peptides. Western Blot, IP, FACS. Standard molecular biology techniques including DNA manipulation and sequencing, polymerase chain reaction (PCR), RT-PCR and real-time PCR. Imaging techniques including in situ hybridization, immunohistochemistry, immunofluorescence, and confocal microscopy. Use of human cell cultures and bacteria.

 

Computer Skills

Familiar with bioinformatics programmes including computer based searches, as well as sequence retrieval and analysis systems. Familiar with PDQuest, and Proteomweaver software for 2D-Gel analysis. MASCOT database search engine. PANTHER expression data analysis. Familiar with MatLab.

Research

Research Interests

My current research focuses on how tumour cells escape stress signals and why they are growing quickly. By identifying the underlying mechanisms we hope to be able to inhibit tumour growth.

·         Raf signalling and apoptosis in Cancer

·         Raf signalling and Alternative Splicing in Cancer

-          We are interested in how alternative splice isoforms of A-Raf kinase contribute to signalling and why they are de-regulated in tumour cells.

·         Spatial regulation of Raf kinases and signalling

-          Raf kinases are found at different localisations within a cell. We want to understand what kind of specific function is associated with this spatial regulation of signalling.

·         Protein expresion noise and signalling

-          Protein expression noise in combination with specific signalling behaviour determines cell fate decisions.
