Biography

Helen Roche's academic background is in Human Nutrition & Molecular Medicine. Her appointment as Associate Professor of Nutrigenomics bridges disciplines based at the UCD Conway Institute and the UCD Institute of Food & Health within UCD. She is also a Science Foundation Ireland (SFI) Principal Investigator and a Food for Health Ireland (FHI) Principal Investigator. In 2014 she won significant funding from the Department of Agriculture, Food and the Marine; NUTRIMAL is a large project grant addressing 'Novel Nutritional Solutions to Combat Malnutrition in the Elderly' in collaboration with the HSE, SafeFood and a number of industry partners. In collaboration with UCC APC, another FIRM award ImmunoMet will address the interactions between nutritional status, metabolic health and the gut microbiome.  Earlier in her career as Wellcome Trust Fellow & Senior Lecturer in Molecular Nutrition at TCD, Dr Roche established the first Nutrigenomics research group in Ireland, at the Institute of Molecular Medicine at TCD.
 
Prof Roche was recently appointed to the Scientific Advisory Board (SAB) of the European Healthy Life Healthy Diet (HDHL) Joint Programming Initiative (JPI). The objective of which is to achieve greater pan-European research integration relating to Food and Health. She as has since been elected chair elect of the HDHL JPI SAB in 2014.   

Nutrigenomics uses state-of-the-art ¿omoics' technologies to investigate the interaction between Nutrition and the Human Genome and gain a greater understanding of the molecular effects of nutrition on health. Whilst it is known that nutrition plays an important role in health and disease, often the cellular and molecular effects of nutrients are not fully understood. In light of the Human Genome Project and the rapid advances in molecular biology, it is essential that these opportunities be applied to nutrition research to progress our understanding of the role of nutrition in health and disease. The potential scope of research within the context of Nutrigenomics is enormous and could include:

¿ Personalised Nutrition & Health - Genetic determinants of nutrient status, metabolic response and predisposition to diet-related diseases (obesity and insulin resistance, type 2 diabetes (T2D), cardiovascular disease (CVD), some chronic pro-inflammatory conditions and some cancers).

¿ Molecular Nutrition - Nutrient regulation of gene / protein expression, genetic determinant of responsiveness to nutritional therapy and/or diet-related disease progression, and the effects of dietary fatty acids on the expression of the genes involved in metabolic health (T2D, CVD, inflammation and colon cancer).

¿ Targeted Nutrition / Functional Food Development - Identifying novel food derived bioactives which have positive effects on molecular processes related to attenuating risk factors associated with T2D, CVD, sub-acute chronic inflammation and insulin resistance.

Research

Research Interests

The SFI Principal Investigator Programme entitled 'Dietary fatty acids: impact on adipose inflammation & insulin resistance - novel therapeutic targets' is determining the interaction between inflammatory and nutrient stressors within the context of obesity, insulin resistance and type 2 diabetes mellitus (T2DM). This project is in collaboration with Prof Kingston Mills, School of Biochemistry & Immunology, Trinity College Dublin, Ireland (http://www.tcd.ie/Biochemistry/research/k_mills.php); Prof Muredach ReillyUniversity of Pennsylvania Cardiovascular Institute (http://www.med.upenn.edu/apps/faculty/index.php/g5165284/p8218) and Prof Jose Ordovas Jean Mayer USDA Human Nutrition Research Center on Aging at Tufts University (http://hnrca.tufts.edu/people/scientists/k-p/jose-m-ordovas-ph-d/). The basic science within our SFI-PI programme compliments other on-going and previous translational projects including, FHI, LIPGENE an EU FP6 IP; NuGO an EU FP6 Network of Excellence and MECHE which focus on the effect of genetic background and nutrient exposure, specifically the molecular effect of dietary fatty acids, on insulin sensitivity and inflammation, within the context of the metabolic syndrome. 


 

Wellcome Trust Career Development Fellowship awarded to Dr Fiona McGillicuddy, supported by Professor Helen Roche as mentor, will determine the ¿Functional consequences of obesity induced adipose tissue inflammation on HDL receptor capacity & reverse cholesterol metabolism'. It provides a novel perspective on the critical interactions between obesity and CVD risk. The Wellcome Fellowship represents an important career development mechanism wherein excellent young scientists gain the opportunity to establish themselves as independent scientists.   
 
In 2014 she won significant funding from the Department of Agriculture, Food and the Marine; NUTRIMAL is a large project grant addressing 'Novel Nutritional Solutions to Combat Malnutrition in the Elderly' in collaboration with the HSE, SafeFood and a number of industry partners. In collaboration with UCC APC, another FIRM award ImmunoMet will address the interactions between nutritional status, metabolic health and the gut microbiome.   

Food for Health Ireland (FHI): This functional foods programme is focused on identifying novel bioactives derived from milk. The role of the Nutrigenomics team within FHI draws upon our high-level expertise in the obesity, insulin resistance, T2D area. It represents the application of skills supported and advanced under our SFI-PI programme, spanning from detailed insight of molecular mechanisms to nutrition and metabolism with a view to providing very comprehensive understanding of putative health effects of novel milk derived bioactives. http://www.fhi.ie/default.asp

MECHE (Metabolic Challenge Study): This translational programme funded by The Food Institutional Research Measure of Department of Agriculture Fisheries and Food has defined the metabolic phenotype of a large cohort of healthy volunteers in response to several stressors including lipid and glucose tolerance tests (OGTT, OLTT) and exercise.  Highly detailed metabolic analysis as well as very detailed nutrigenomics profiling utilising transcriptomics, proteomics and metabolomics provides a rich data source. It is a sub-component of the larger JINGO (Joint Irish Nutrigenomics Organisation) initiative led by Professor Mike Gibney http://www.ucd.ie/jingo/

LIPGENE: Diet, genomics and the metabolic syndrome: an integrated nutrition, agro-food, social and economic analysis. LIPGENE is an EU Framework 6 Integrated Project designed to determine the effects of dietary fatty acid composition and human genetic variation on the development and risk of the metabolic syndrome. Project involves a prospective genetic study and a human dietary intervention trial to test the interaction between dietary fat exposure and genetic risk of the metabolic syndrome. HRB Molecular Medicine PhD : The Nutrigenomics Research Group are part of the HRB funded PhD Trining Programme in Molecular Medicine based at Trinity College Dublin. NutSysBiol: Nutrigenomics & Functional Food Development: Towards a Nutritional Systems Biology Approach. This project will test the robustness of transcriptomic and proteomic approaches in determining the heath effects of fatty acid modified functional foods. A nutritional systems biology approach will be developed to understand the role of nutrients designed to prevent diabetes and atherosclerosis. This project is in collaboration with Dr Aidan Moloney, Teagasc Ashtown Food Research Centre, Ireland (http://www.teagasc.ie) and Dr Baukje De Roos, Rowett Research Institute, Aberdeen, Scotland (http://www.rowett.ac.uk/divisions/vhp/b_deroos.html). The Nutritional Systems Biology Programme is also being supported by IRSCET who are funding a PhD Programme in Nutritional Systems Biology within their EMBARK Postgraduate Research Scholarship Scheme (http://www.ircset.ie).

Functional Foods: Healthy fatty acid-enriched fresh beef: Implications for shelf-life, flavour and the health of the consumer. The project will determine if the health effects of CLA enriched beef are as potent as synthetic forms of the fatty acid on insulin sensitivity in male ob/ob mice, atherosclerosis in apo E knock out mice and chronic inflammation in a mouse model of IBD. This project is part of a larger FIRM programme being co-ordinated by Dr Aidan Moloney, Teagasc Ashtown Food Research Centre and is in collaboration with Dr Frank Monaghan, UCD School of Agriculture, Food Science & Veterinary Medicine (http://www.ucd.ie/agfoodvet/index.html).

Clare Reynolds & Jolene Mc Monagle: Functional Foods 'Healthy fatty acid-enriched fresh beef: Implications for shelf-life, flavour and the health of the consumer'. The project will determine if the health effects of CLA enriched beef are as potent as synthetic forms of the fatty acid on insulin sensitivity in male ob/ob mice, atherosclerosis in apo E knock out mice and chronic inflammation in a mouse model of IBD.

Elizabeth Oliver & Emma Allot: HRB PhD Mol Med The Nutrigenomics Research Group are part of the HRB funded PhD Trining Programme in Molecular Medicine based at Trinity College Dublin. This programme is supporting two PhD Research Programmes entitled ‘The role of the adipocyte in insulin resistance' and ‘The molecular interactions between Obesity in Cancer'.

Enhancement of Teaching

Molecular Nutrition (Hum Nut 40200) is a very new module associated with the BSc Human Nutrition - nevertheless it is important to continuously review to enhance the curriculum. In order to ensure that BSc is attaining the Programme Learning Outcomes targets it set out to achieve - Geraldine O¿Neill (UCD Teaching & Learning) facilitated a curricular review in November 2012. In particular group structure assessment methodologies have been set in place. Overall this process ensured that our curriculum was attaining all teaching and learning objectives we set out to achieve. The forms of continuous assessment used within the BSc Human Nutrition / Molecular Medicine Module at UCD very progressive and focus particularly on critical evaluation, problem solving and presentation skills. In the Molecular Nutrition module the students are introduced to a ¿Commentary Style¿ critique of high impact papers. To this end, as part of their continuous assessment each student must complete a one page commentary on a topical, high impact paper published in the last 5 years ¿ in a given subject area that compliments preceding lectures. Added to this the student present an 'Elevator Pitch' / 'Hot Topic' style presentation, which consists of 4-5 slides within 10 minutes with 5 minutes Q&A, to their class which presents the main content of their commentary / critique they completed of the paper they reviewed. This exercise enhances both written and oral skills for problem solving and critical assessment.

Review and Enhancement of Curriculum

Molecular Nutrition is a very new approach to within the field of nutrition, therefore there are no classical textbooks dedicated to this specific subject matter. Students can refer to the Nutrition & Metabolism Textbook series Molecular Nutrition Chapter as a reference material related to this module. Nevertheless the students have a high reliance on the lecture material. Given the lack of classical textbook material I take a lot of time to select and present the most important papers relevant to the subject matter discussed in each lecture. This additional reading material is always made available via Blackboard as often some of the papers / journals require payments. Working through such papers trains the student to develop a critical eye in terms of assessing the merits and limitations of different experimental approaches.

Developing as a Teacher

Translating research excellence into advanced Teaching and Learning has always been a priority in order to harness the full assets of research within the academic environment. To this end I have spear-headed the new Molecular Nutrition Module at UCD. This is a highly topical and rapidly advancing state-of-the-art subject area - therefore most of the material is derived from high-impact peer review publications. I have developed as a teacher in terms of enhancing the breath and focus of nutrition as a discipline. I was trained initially in a clinical context with a strong focus on metabolism and health, to which I added strong research and molecular perspectives. To this end, I have I have taken the opportunity to develop and implement a number of state-of-the-art perspectives within the BSc Human Nutrition, Masters in Public Health and the Conway Postgraduate Modules which broaden the opportunities for greater multi-disciplinary perspectives to human nutrition.
