Research Interests
| Research in our group focuses on the structure, function and regulation of monoamine neurotransmitter transporters, in particular the serotonin transporter (SERT). SERT is an integral plasma membrane protein that functions in the high-affinity re-uptake of the neurotransmitter serotonin (5-hydroxytryptamine, 5-HT). SERT belongs to a large family of sodium-dependent neurotransmitter transporters. SERT is the primary target for a number of widely prescribed antidepressant drugs, including tricyclic antidepressants (TCA) such as imipramine and selective serotonin re-uptake inhibitors (SSRI) such as citalopram, fluoxetine (Prozac) and paroxetine. Multiple pathways have been implicated in neurotransmitter transporter regulation. Functional activity as well as subcellular localisation and targeting of SERT and related transporters are thought to depend on the interaction with specific regulatory proteins. One of the major programmes in our group is the identification and characterization of SERT-interacting proteins using various methods (yeast two-hybrid method, co-immunoprecipitation, confocal microscopy and proteomics approaches), and to study the dynamics of these interactions in response to extracellular stimuli and to the activation of various signaling pathways. For example, we currently study SERT regulation by interacting proteins in blood platelets during platelet activation. Another major project concerns the regulation of SERT gene expression by pro-inflammatory cytokines. Neurotransmitter transporters also appear to be regulated by protein-lipid interactions. We recently found that SERT localises to lipid microdomains, so-called rafts (Magnani et al., 2004, JBC 279:38770-8). Current research in our group focuses on the elucidation of the mechanism by which lipid rafts affect SERT function and on the identification of proteins that associate with SERT in lipid microdomains
