Chromatography

In January 2022 our Agilent 1200 HPLC (High Pressure Liquid Chromatography) instrumentation suite was upgraded, with the addition of new Agilent 1290 UHPLC (Ultra High-Pressure Liquid Chromatography). The purchase of the UHPLC was made possible through the UCD EQUIP Scheme.

The Chromatography suit is open to SFI and non-SFI researchers from the school and externally, to industry, and other national and international users.

The laboratory also includes an Agilent 8860 GC (gas chromatography) instrument.

INSTRUMENTATION
Reverse Phase UHPLC Setup:

This is an Agilent 1290 system with a 1290 Flexible quaternary pump (G7104A) with an operating pressure of 1300 bar. It has a maximum operating flow rate of 5mL/min and a minimum flow rate of 0.01mL / min adjustable in increments of 0.01 mL/min. The minimum solvent gradient is 0.1% (if using more than one channel). This pump also contains an inline degasser to remove trace dissolved gases.

The Agilent 1290 Infinity II ALS (automated liquid sampler) a G7167A can hold 336 over 6 sample trays of standard 2 mL vials with a maximum injection volume of 20 uL in increments of 0.1 uL with a minimum injection volume of 0.1uL. The error in the injection volume is 10 nL (or 0.15% via standard deviation) with an injection linearity of 0.9999 within 0.1 uL to 20 uL. The sample trays can be removed unused with 96 well plates or 384 well plates. The autosampler can also be temperature controlled with the temperature range of 4 °C to 40 °C in increments of 1 °C.

There is a 6-position 1290 MCT (multicolumn thermostat) which is a G7116B capable of holding 6 columns with a maximum operating temperature of 60 °C adjustable in increments of 1 °C. Just allows for rapid column changing during the method development.

The 1290 Infinity II VWD (variable wavelength detector) is a G7114B capable of capturing a single wavelength at 240 Hz or 120Hz at jewel wavelength detection. It's a double beam photoemitter containing a deuterium lamp with a wavelength range of 190 to 600 nm. Wavelength accuracy within a nanometer self-calibrating with deuterium lines verification with holmium oxide filter. Wavelength precision is within 0.1 nm.

The Agilent 1260 Infinity II FLD (fluorescence detector) attached to this instrument is a G7121A which is capable of single wavelength (excitation and emission). The detector has a xenon flash lamp with the maximum data sampling rate of 74 Hz. The excitation monochromator has a range of 200 to 1200 nm with a fixed bandwidth of 20 nm. the emission monochromator has a range of 200 to 1200 nm with the 20 nm fixed bandwidth.

There is also an Agilent 1290 Infinity II RID (Refractive index detector) which is a G7162B which has a maximum operating range of 600 x 106 RIU with a maximum data sampling frequency of 148 Hz. This detector also has a solvent recycling function that allows you to recycle your element to save on solvent costs.

Normal Phase UHPLC Setup:

There is also an Agilent 1260 Infinity II DAD (Diode Array Detector) which is a G7115A attached to this instrument with deuterium and tungsten lamps capable of operating wavelength range of 190 to 950 nm. Wavelength accuracy within a nanometer self-calibrating with deuterium lines verification with holmium oxide filter. Wavelength precision is within 0.1 nm with programmable slit widths of 1, 2, 4, 8 and 16 nm. It has a maximum sampling rate of 120 Hz.

Gas Chromatography setup:

An Agilent 8860 GC equipped with an FID (flame ionising detector) and a 50-position automated liquid sampler. The instrument is set up that split/splitless injections are possible, and our carrier gas is helium.

Reverse Phase HPLC Setup:

This is an Agilent 1200 series system with a binary pump (G1312A) with the max operating pressure of 400 bar. The minimum solvent gradient is 0.1% (if using more than one channel) with a maximum flow rate of 5 mL/min on a minimum flow rate of 0.1 mL/min in adjustable increments of 0.1 mL/min. The ALS (automated liquid sampler) is a G1329A which can hold up to 100 samples (2 mL sample vials) with maximum injection volume of 100 uL in adjustable increments of 0.1 uL with a minimum injection volume of 0.1uL. There is a six-position column switching valve attached to the system for rapid column changing and method development and thermostatically controlled column department (G1316A). There is a DAD detector is a (G1315D) capable of measuring eight different wavelengths at 80Hz or full spectral data at lower resolution. Also Attached is an RID (refractive index detector) is a G1362A which can be used to detect molecules that have no UV/vis active regions.

400 bar. The minimum solvent gradient is 0.1% (if using more than one channel) what's the maximum flow rate of 5 mL/min on a minimum flow rate of 0.1 mL/min in adjustable increments of 0.1 mL/min. 

The ALS (automated liquid sampler) is a G1329A which can hold up to 100 samples (2 mL sample vials) with maximum injection volume of 100 uL in adjustable increments of 0.1 uL with a minimum injection volume of 0.1uL. There is a six-position column switching valve attached to the system for rapid column changing and method development and thermostatically controlled column department (G1316A). There is a DAD detector is a G1315D) capable of measuring eight different wavelengths at 80Hz or full spectral data at lower resolution.
